KLOW is a four-peptide complex studied in the field of tissue regeneration, skin condition, and inflammation regulation. To the GLOW formula (the copper peptide GHK-Cu, BPC-157, and TB-500), it adds KPV, a tripeptide derived from the α-MSH hormone that is being researched for its anti-inflammatory action. Each component acts on a different phase of repair: collagen synthesis, blood vessel formation, cell migration, and inflammation control. What is KLOW (GHK-Cu + TB-500 + BPC-157 + KPV)

KLOW combines four peptides that have been studied separately:

  • GHK-Cu (50 mg): a complex of the tripeptide Gly-His-Lys with copper, naturally present in human plasma.
  • BPC-157 (10 mg): a synthetic 15-amino acid peptide whose sequence is derived from a protein in gastric juice.
  • TB-500 (10 mg): a synthetic peptide based on the active region of thymosin beta-4.
  • KPV (10 mg): the tripeptide Lys-Pro-Val, the C-terminal end of the alpha-melanocyte-stimulating hormone (α-MSH).

GHK-Cu is the major component and provides the line of study on the extracellular matrix and the skin. BPC-157 and TB-500 provide tissue repair, and KPV provides inflammation regulation. Together, the formula covers the main phases of tissue repair: inflammation, blood vessel formation, cell migration, and remodeling.

What do the studies show

KPV (Lys-Pro-Val) corresponds to the last three amino acids of α-MSH, the region responsible for much of its anti-inflammatory activity. Unlike the full hormone, KPV is not associated with skin pigmentation.

Its best-described mechanism is intracellular: it enters cells via the peptide transporter PepT1 and, once inside, reduces NF-κB pathway activation and the production of proinflammatory cytokines. In cellular and animal models, it has been studied in relation to:

  • intestinal inflammation in colitis models (Dalmasso et al., Gastroenterology, 2008);
  • skin inflammation and wound healing;
  • the antimicrobial activity described for the C-terminal fragments of α-MSH.

Its molecular formula is C16H30N4O4 and its molecular weight is 342.4 g/mol.

Laboratory application

The other three components of KLOW have their own lines of research:

  • GHK-Cu: in fibroblasts, it stimulates the synthesis of collagen, elastin, and glycosaminoglycans, and regulates the enzymes that remodel the extracellular matrix. This is why copper peptides are also a common ingredient in cosmetics.
  • BPC-157: studied primarily in rodents, it is associated with the formation of new blood vessels through VEGF and nitric oxide, as well as the repair of tendons, ligaments, and digestive mucosa.
  • TB-500: replicates the region of thymosin beta-4 that binds to actin. Tβ4 has been studied for its role in cell migration and in the repair of wounds in the skin, cornea, and heart.

Each peptide has been researched separately. The interest in combining them lies in the fact that their mechanisms complement each other without overlapping.

Format and order

KLOW is supplied in a pen, a ready-to-use format that does not require the reconstitution of a lyophilisate. It contains 80 mg of peptides in 3 ml of solution, with the following concentrations:

  • GHK-Cu: 50 mg (16.7 mg/ml);
  • BPC-157: 10 mg (3.3 mg/ml);
  • TB-500: 10 mg (3.3 mg/ml);
  • KPV: 10 mg (3.3 mg/ml).

The scale has 300 clicks of 0.01 ml. Due to the copper in the GHK-Cu, the solution has a characteristic blue color. The pen should be stored in a refrigerator, between 2 and 8 °C, without freezing and protected from direct light.
