Epitalon, also spelled Epithalon, is a synthetic tetrapeptide composed of four amino acids: alanine, glutamic acid, aspartic acid, and glycine (Ala-Glu-Asp-Gly, or AEDG). It was developed at the St. Petersburg Institute of Bioregulation and Gerontology under the direction of Vladimir Khavinson. It is the synthetic version of epithalamin, a peptide extract from the pineal gland.

Epithalamin is a mixture of peptides; Epitalon, by contrast, is a single molecule with a known sequence. Its small size and stable structure facilitate the reproducibility of experiments.

Its CAS number is 307297-39-8, its molecular formula is C14H22N4O9, and its molecular weight is 390.35 g/mol.

Telomeres are the protective ends of chromosomes and shorten with each cell division. Telomerase is the enzyme capable of lengthening them. In a study with human fibroblast cultures, Khavinson et al. (Bulletin of Experimental Biology and Medicine, 2003) observed that Epitalon induced telomerase activity and telomere elongation.

In animal models, Epitalon has been studied in relation to:

aging biomarkers and lifespan in rodents (Anisimov et al., Biogerontology, 2003);
the incidence of spontaneous tumors in aging mice;
aging processes and cognitive functions.

Most of the research on Epitalon comes from Vladimir Khavinson's group in St. Petersburg, which developed the peptide.

The pineal gland regulates circadian rhythms through melatonin, whose nocturnal production decreases with age. Epitalon was designed from an extract of this gland, and much of its research focuses on pineal function:

the restoration of the nocturnal melatonin peak in aged rhesus monkeys (Khavinson, Goncharova, and Lapin, Neuroendocrinology Letters, 2001);
daily rhythms of norepinephrine and dopamine in the hypothalamus, in animal studies;
age-related neuroendocrine regulation.

Due to this relationship with melatonin and daily rhythms, Epitalon is also a subject of study in chronobiology.

Epitalon is supplied as a spray, a ready-to-use format that does not require the reconstitution of a lyophilized powder.
