What is BPC-157 + TB-500 (Wolverine stack)

The complex combines two distinct classes of molecules that, in preclinical models of injury, act at different stages of tissue repair.

BPC-157 is a synthetic 15-amino acid peptide (Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val). Its sequence is derived from a protein found in human gastric juice called BPC (body protection compound). Its CAS number is 137525-51-0 and its molecular weight is approximately 1419 g/mol.

TB-500 is a synthetic peptide based on thymosin beta-4 (Tβ4), a 43-amino acid protein found in almost all cells of the body. It replicates its active region, the one that binds to actin.

Each peptide has been investigated separately. The interest in combining them is based on the fact that their mechanisms are complementary: BPC-157 acts primarily at the vascular and connective tissue level, and TB-500 on the migration of cells to the damaged area.

What do the studies show

In experimental models, mainly in rodents, BPC-157 has been studied in the following areas:

  • increase of angiogenic factors, such as VEGF, and the expression of its receptor VEGFR2;
  • participation in nitric oxide (NO)-dependent vascular responses;
  • modulation of inflammatory mediators, such as the NF-κB pathway and pro-inflammatory cytokines;
  • repair of the digestive tract mucosa;
  • healing of tendons and ligaments, with increased fibroblast proliferation and migration.

Much of this work comes from Predrag Sikirić's group at the University of Zagreb. Among the most frequently cited studies are those by Chang et al. (Journal of Applied Physiology, 2011), on tendon fibroblasts, and Hsieh et al. (Journal of Molecular Medicine, 2017), on VEGFR2 activation.

Laboratory application

Most of the published research has been conducted with full-length thymosin beta-4, whose active region is replicated by TB-500. In cellular and animal models, it has been studied in relation to:

  • actin regulation: Tβ4 is the primary protein that sequesters monomeric actin in the cell, which influences its shape and movement;
  • cell migration to the damaged area;
  • angiogenesis;
  • skin, corneal, and cardiac tissue wound repair.

Reference works include Malinda et al. (Journal of Investigative Dermatology, 1999), on skin healing, and Bock-Marquette et al. (Nature, 2004), on cardiac repair in mice.

Format and order
