What do the studies show

In addition to participating in oxidation-reduction reactions, NAD+ is the cosubstrate for several groups of enzymes that consume it as they act:

  • Sirtuins (SIRT1–SIRT7): deacetylases that regulate gene expression, metabolism, and the stress response. Their activity depends directly on NAD+ availability.
  • PARPs (poly-ADP-ribose polymerases): enzymes involved in DNA repair.
  • CD38: an enzyme that degrades NAD+ and whose activity increases with age in animal models.

In the mitochondria, NADH donates its electrons to complex I of the respiratory chain, an essential step for ATP production. For this reason, the balance between NAD+ and NADH is considered an indicator of the cell's energy status.

Laboratory application

Research on NAD+ focuses on several lines:

  • the decline of NAD+ levels with age, observed in animal models and in some human tissues;
  • the relationship between that decline and mitochondrial function;
  • the role of sirtuins in metabolism and cellular aging;
  • strategies to restore NAD+ levels, using the molecule itself or its precursors, such as NMN or NR.

Key reference works include Gomes et al. (Cell, 2013), which linked the decline of NAD+ to impaired communication between the nucleus and the mitochondria in aged mice. Also, the study by Camacho-Pereira et al. (Cell Metabolism, 2016), which identified CD38 as one of the main causes of that decline, and Verdin’s review (Science, 2015) on NAD+ in aging, metabolism, and neurodegeneration.
