MOTS-c is a mitochondria-derived peptide: its sequence is encoded in the mitochondrial DNA, within the MT-RNR1 gene, and not in the cell nucleus. It belongs to a relatively recently described family of signaling molecules, the mitochondria-derived peptides. What do the studies show

The best-described mechanism of MOTS-c focuses on skeletal muscle:

  • it inhibits the folate cycle and de novo purine synthesis;
  • this causes the accumulation of AICAR, a metabolic intermediate;
  • AICAR activates AMPK, the enzyme that acts as the cell's energy sensor and regulates glucose uptake and fat oxidation.

In situations of metabolic stress, MOTS-c translocates to the cell nucleus. There, it interacts with the NRF2 factor and regulates the expression of antioxidant response genes (Kim et al., Cell Metabolism, 2018). It is a direct example of communication between the mitochondria and the nucleus.

In laboratory models, MOTS-C is investigated across several avenues of research:

  • AMPK activation, a key enzyme regulating cellular energy metabolism;

What is MOTS-c?
